原花青素与珍珠粉混合物对老龄大鼠肝细胞、线粒体酶及果蝇生存期的影响

摘要 目的 通过对老龄大鼠肝细胞及线粒体酶活性、果蝇生存期的检测 ,探讨原花青素与珍珠粉混合物延缓衰老的作用机制。方法 以原花青素与珍珠粉混合物为受试物 ,将 2 0月龄SD大鼠随机分为空白对照组和 0 1g/ (kg .bw) (低 )、0 2g/ (kg.bw) (中 )、0 6g/ (kg .bw) (高 ) 3个剂量组 ,每组 11只动物 ,连续灌胃 35d ,取肝脏 ,测定肝细胞及线粒体超氧化物歧化酶 (SOD)、谷胱苷肽过氧化物酶 (GSH Px)及丙二醛 (MDA)的含量。将OregonK野生型黑腹果蝇随机分为空白对照组和 0 0 0 7%、0 0 13%、0 0 4 0 %、0 12 0 %剂量组 ,每组 4 0 0只 ,雌雄各半 ,隔天一次统计果蝇存活数、死亡数 ,直到全部死亡 ,计算平均寿命、最高寿命及半数死亡时间。结果 原花青素与珍珠粉混合物各剂量组均能升高老龄大鼠肝细胞线粒体GSH Px活性 (P <0 0 5或P <0 0 1) ,中、高剂量组能升高SOD活性 (P <0 0 5或P <0 0 1)和降低MDA含量 (P <0 0 5或P <0 0 1) ;受试物亦能升高老龄大鼠肝细胞SOD活性和降低MDA含量 (均P <0 0 1) ;果蝇生存试验中 ,受试物各剂量组的半数死亡时间比对照组延长 2~8d ,且明显延长果蝇的平均寿命和最高寿命。结论 原花青素与珍珠粉混合物可能通过提高肝细胞及线粒体GSH Px、SOD活性 。 Objective To explore the mechanism of delaying aging of GSPE (Grape Seed Extract Proanthocyanidin) and pearl powder mixture. Methods (1) 20-month-old SD rats were randomly divided into four groups (n=11 each). The mixture was injected to stomach with the dosage of 0 g/kg, 0.1 g/kg, 0.2 g/kg and 0.6 g/(kg.bw) for thirty-five days, SOD, GSH-Px activity and MDA content were measured. (2) Oregon K flies were divided into one controlled group and four tested groups (n=400 each) with the dosage of 0%, 0.007%, 0.013%, 0.040% and 0.120%. Counted the number of live and death every other day until all the flies died and calculate the mean and maximum life-span and half-death time of fruit flies. Results GSPE and pearl powder mixture could significantly increase GSH-Px activity of liver mitochondria in three dose groups, increase SOD activity in the middle and high dose groups and decrease MDA content. It could significantly prolong the mean and maximum life-span and half-death time of fruit flies also. Conclusion GSPE and pearl powder mixture have the effect of delaying ageing by increasing enzymes activity of hepatocyte and liver mitochondria and reducing generation of oxygen free radicals.
