摘要
目的研究—氧化氮合酶(NOS)表达与肝癌增殖凋亡的关系。方法用免疫组化的方法对21例肝癌及癌旁组织中的诱导型一氧化氮合酶(iNOS)及增殖细胞核抗原(PCNA)的表达进行原位检测和观察,并与脱氧核糖核酸末端转移酶介导的缺口末端标记(TUNEL)的凋亡细胞进行比较。结果癌旁组织 iNOS 阳性,非癌组织多阴性或弥漫弱阳性;iNOS 在周边的癌组织及侵入纤维组织中的癌细胞阳性,癌组织核心多阴性或弥漫弱阳性。PCNA 表达与 iNOS 相似,iNOS 表达阳性区 PCNA 表达阳性率也高。TUNEL 染色凋亡细胞阳性率高区也是 PCNA 阳性率高区。结论 iNOS 表达与肝癌的增殖凋亡有关,在肝癌的发生发展中起重要作用。
This study was to investigate the relation of nitric oxide synthase with cell proliferation and apoptosis in hepatocellular carcinoma(HCC).Methods The expression and localization of inducible nitric oxide synthase (iNOS)and PCNA in the 21 patients with HCC were studied by immunohistochemistry.The apoptosis in tissue section was detection by TUNEL(terminal deoxynucleotidyl transferase[TdT]-mediated desoxyuridinetriphosphate[dUTP]nick end-labeling)Results The iNOS protein was markedly expression at the liver tissue of hepatic cirrhosis or chronic liver hepatitis,which was close to tumor edge.Meanwhile it was weakly or undetectable at the liver tissue localized beyond 1. 5cm from the tumor edge.The iNOS protein was markedly expression at the invasive tumor edge and HCC cells that inva- sived into stroma,but it was weakly or undetectable at the most tumor core.The expression of iNOS was related to an in- creased proliferative activity of cells,and moreover,the apoptotic cells was related to PCNA positive cells(p<0.01). ONCLUSION Excessive expression of iNOS may contribute to the progress of HCC.
