中国HIV-1感染人群和普通人群四种基因多态性分布的差异分析 被引量:3

An analysis of the difference between four gene polymorphism of HIV-1 positive Chinese and general Chinese populations

摘要 目的 艾滋病病毒 (HIV) 1型感染辅助受体CCR5、CCR2和SDF 1等基因的多态生与HIV 1感染和发病过程有密切的关系 ,但上述基因对中国人群易感性的影响尚不清楚 ,为此 ,研究了中国HIV 1感染者四种基因型的分布特点 ,以了解其对中国人群HIV 1易感性的影响。方法 HIV 1感染者被分为经血液传播途径感染和经性传播途径感染的两类人群 ,应用PCR RFLP技术分析HIV感染者CCR5Δ32、CCR2 6 4I、SDF1 3′A、CCR5m30 3四种基因多态性 ,使用SPSS11 0统计分析软件分析HIV感染者与中国正常人群上述基因型分布的差异。结果 在189例HIV 1感染者中 ,没有发现CCR5Δ32和CCR5 m30 3突变基因型 ;SDF1 3′A等位基因突变频率为 2 6 14 % ,CCR2 6 4I的突变频率为 19 82 %。χ2 检验的结果发现 ,中国HIV 1感染者SDF1 3′A、CCR2 6 4I基因多态性分布与中国正常人群没有显著性差异 (χ2 =1 2 6 6、0 138,P值均 >0 5 )。对血液传播途径感染人群和性传播途径感人群分别进行统计 ,同样发现与中国正常人群SDF1 3′A、CCR2 6 4I基因型的分布没有显著性差异。结论 SDF1 3′A、CCR2 6 4I基因多态性突变 ,对于中国人群HIV 1X经血液途径和性途径的感染没有明显的阻断作用。 Objective Recent researches show HIV-1 infection coreceptor(CCR5Δ32,CCR2-64I,SDF1-3'A,CCR5m303CCR5,CCR2 and SDF-1)gene polymorphism have close relation with human HIV-1 infection,but it is unknown how the above gene polymorphism affect Chinese HIV-1 susceptibility.Thus CCR5Δ32,CCR5m303,CCR2-64I,SDF1-3'A gene polymorphism of Chinese HIV-1 carriers were analyzed to find the relation of above gene polymorphism with Chinese HIV-1 susceptibility.Methods A11 the HIV-1 carriers were divided into two groups:77 persons (through blood transmission)and 83 persons (through sexual transmission).PCR/RFLP technology was used to analyze CCR5Δ32,CCR2-64I,SDF1-3'A and CCR5m303 gene polymorphism.SPSS11.0 software was used to analyze the diversity between Chinese general population and HIV-1 infectors.Results There were not CCr5Δ32 and CCR5m303 mutation genotype in 171 HIV-1 infectors.SDF1-3'A allele frequency is 26.14,CCR2-64I allele frequency is 19.82.The result of Chi-square Tests show there are no significant diversity between Chinese general population and HIV-1 carriers in SDF1-3'A and CCR2-64I genotype distribution(χ2=1.266?0.138,P>0.5).There are norther no CCR2-64I genotype distributing(χ2=1.266?0.138,P>0.5).There are also no significant diversity between Chinese general population and HIV-1 carriers who infected HIV-1 through blood transmission or through sexual transmission in SDF1-3'A and CCR2-64I genotype distribution.Conclusion SDF1-3'A,CCR2-64I gene polymorphism mutation do not have effect on Chinese HIV-1 susceptibility.
