摘要
利用聚丙烯酰胺凝胶电泳,对中华大蟾蜍(Bufo bufo比gargarizans)与花背蟾蜍(Bufo raddei)种间核移植胚胎发育早期六个不同阶段中全胚胎的乳酸脱氢酶(LDH)同工酶进行了分析。酶谱比较的结果表明:在正、反种间移核胚胎中,供体核LDH基因的活动开始表现于尾芽胚期;此前,杂种胚胎中LDH同工酶谱类型与受体一致。
Using polyacrylamide gel electrophoresis, enzyme phenotype of lactate dehydrogenase (LDH) in nucleo-cytoplasmic hybrids obtained through nuclear transplantation between Bu-fo bufo gargarizans and Bufo raddei was investigated in different stages of early development of the nuclear transplanted embryos. In two aspects the experiment was implemented to demonstrate the cytoplasmic influence on the expression of foreign nuclear activity , and the interelation between the arresting of development and nucleo-cytoplasmic incompatibility. During the experiment, in general, the developmental anomalies were observed just from the start of gastrulation, and from then on the nucleo-cytoplasmic hybrids exhibited several anomalies, which developed no further than the heart-beating stage.In Genus Bufo, LDH isozymes showed differences in electrophoretic mobility which was connected with specific types. In the cases happened in Bufo bufo gargarizans and Bufo raddei, the natural marks for identifing the form of zymograms in the early development of nucleo-cytoplasmic hybrid embryos were constituted. Electrophoretic zymograms revealed the grafted nuclei activity from stage 17(tail bud stage), before which the LDH isozymes are provided by the recipient cytoplasm in both species (host). However, the fractions corresponding to initial cytoplasmic LDH isozymes of host lasted until the embryos died. In addition , we also observed that the quantity of LDH bands on the gel of grafted nucleus activity increased relatively, while the staining of the same bands corresponding to LDH of the initial egg cytoplasm reduced. Those indicated that, with development proceeding, the grafted nuclear effect on LDH isozymes characteristics became prevalent in embryos, if the nucleo-cytoplasmic hybrids develop over tail bud stage.
