摘要 目的 用聚乳酸构建一种具有纵行排列管径的三维立体神经组织工程支架 ,并观察体外培养的雪旺细胞在支架上的生长情况。方法 用组织块培养法获得新生SD大鼠坐骨神经的雪旺细胞 ,利用反复贴壁法纯化雪旺细胞。将消旋聚乳酸R2 0 6采用热致相分离法制备成三维立体支架 ,并涂俯 5 0 μg ml浓度的纤维粘连蛋白 ,把雪旺细胞种植到该支架材料上培养 2周 ,扫描电镜观察。结果 构建的支架材料为白色、泡沫状 ,具有管状定向排列的立体结构 ,直径 60~ 10 0μm ,孔积率为 91%,重均分子量 3 3 0× 10 3。体外培养的雪旺细胞成功种植在该支架上 ,在材料的深处有细胞生长。结论 用热致相分离法制备的聚乳酸三维立体支架有良好的立体构型 ,类似天然周围神经结构。该支架有较好的生物相容性 。 Objective To fabricate polylactide three dimensional scaffolds with oriented porous morphology for injured peripheral nerves and to observe the growth of Schwann cells cultured in vitro on the scaffolds. Methods The resomer (R206) scaffolds were fabricated using a thermally induced polymer solvent phase separation. The foams were incubated with 50 μg/ml solution of fibronectin (Fn) to facilitate strong cell adhesion. The Schwann cells isolated from the newly born SD rat sciatic nerves were seeded onto the scaffolds for 2 weeks. Cell attachment and the morphological changes were observed by scanning electron microscopy (SEM). Results The fabricated scaffolds had three dimensional oriented porous morphology with the diameters from 60 μm to 100 μm, the porosity of 91%, and molecular weight of 330 000. Schwann cells were seeded on the scaffolds successfully. Conclusion The polylactidd scaffolds with oriented porous morphology, similar to the structures of peripheral nerves, have good biocompatibility for the in vitro growth of Schwann cells.
