摘要
目的 :探讨大鼠正常心肌间质胶原纤维网络的形态结构 ,及缺血、缺血 再灌注对其影响。方法 :成年大鼠分成正常组、缺血组、缺血—再灌注组。夹闭左冠状动脉旋支制备缺血模型 ;左心室前壁切取心肌 ,扫描电镜观察。结果 :心肌间质胶原网络由粗、细和微细三种胶原纤维束组成。粗纤维多横行走向 ,环绕多个心肌细胞 ;细纤维于粗纤维深层交织成网 ,跨越相邻心肌细胞 ;微细纤维紧贴心肌细胞表面 ,很少跨越相邻心肌细胞。缺血组使胶原纤维网络破坏 ,再灌注使其破坏加重。结论 :心脏胶原纤维束在形态上分粗、细和微细三种 ,在空间排列上分浅、中、深 3层。缺血、缺血 再灌注均使心肌胶原网络破坏。心肌胶原的破坏可能是影响心功能恢复的原因之一。
Objective: To investigate the architecture and morphology of the collagen network, and probe changes of collagen fibers after ischemia and ischemia reperfusion in rat heart. Methods: Adult rats were randomly divided into normal group, ischemic group and ischemic reperfusion group. Ischemic model was prepaired by occluding circumflex branch of coronary artery. Myocardial block was cut from anterior wall of left ventrical distributed alone circumflex branch of coronary artery. The collagen of myocardial interstitium was observed by scanning electron microscope. Results: myocardial collagen bundle were composed of thick, fibril and microfibril bundle in normal heart. The thick collagen bundle a transversely and revolved around many cardiac muscle cells. The fibril located beneath thick bundle, interwoven with each other, and strided across cells nearly. The microfibril collagen bundle was closely adhered on the cardial cell surface with few cross other cells. The myocardial collagen net was damaged in ischemic heart and seriously damaged in ischemia reperfusion heart. Conclusion: The myocardial collagen fiber bundle can be divided into thick, fibril and microfibril bundle in morphology, and they are arranged in superficial, middle, deep layers in dimension. The myocardial ischemia and ischemia reperfusion all damage myocardial collagen fibers. Damage of myocardial collagen network is one of factors disturbing recovery of cardiac function.
