摘要
作者对自行研制的PCR-100半导体加热和致冷并由微电脑控制的反应装置应用的有效性进行了研究.首先测定PCR-100管内外温度特征曲线,继而对不同来源、不同长度模板DNA进行扩增,对扩增产物以Southern杂交、DNA序列分析验证,结果表明:该仪器性能优良,适用于各种PCR反应.该仪器用于PCR非同位素标记cDNA探针也取得满意结果.
t The applicability and validation of an automatic reaction apparatus (PCR 100) designed by the authors was studied. The apparatus was heated and cooled by semiconductor and controlled by microcomputer. The temper ature property curves of the heat block of the apparatus and samples in microcemnfuge lube in the heat block were tested. Then, the template of genomic DNA, cDNA and plasmid DNA were amplified, respectively. The length of amplified products ranged from 110-1600 base pairs. Moreover, the bio-cDNA probe of epidemic hemorrhagic lever virus was prepared. The results demonstrated that PCR-100 showed desirable performance.
