重组成骨生长肽对同种异体骨移植愈合的实验研究 被引量:4

摘要 目的 观察重组成骨生长肽 (rOGP)对兔同种异体皮质骨移植愈合的影响。方法 采用日本大耳白兔造成桡骨中段 1.0cm缺损植以同种异体骨的动物模型 ,4 2只兔被分成实验组和对照组 ,每组 2 1只。从术后第 1天至取材的前 1天实验组动物每日经耳缘静脉注射rOGP 0 .5 μg/kg ,对照组注射同等剂量的生理盐水。于术后第 4 ,8,12周取材分别行大体、放射线和HE染色组织学观察愈合情况。结果 实验组和对照组组织愈合情况相似 ,实验组有多量的纤维结缔组织包裹连接处 ,放射线观察第 12周骨髓腔通畅及骨改建好于对照组。术后第 8,12周实验组移植骨和宿主骨连接处新生骨小梁多于对照组 (P <0 .0 5 )。 Objective To evaluate the effect of recombinant osteogenic growth peptide(rOGP) on the healing of bone allograft transplanted/implanted in rabbits. Methods Fourty two Japan rabbits were made animal model that 1.0cm bone allografts were transplanted in tibial defect.Fourty two rabbits were divided into experimental and control groups.The rOGP was administered to experimental animals by intravenous injection(0.5μg/kg·d) from the first day after surgery to the day before sacrifice,while the control animals were injected with saline solution.Seven rabbits were killed at 4,8,12 weeks after surgery and obtained samples.The samples were observed with gross,X ray,histological examination(hematoxylin and eosin stain).The histological examination was done quantitative analysis of the callus area amounts of newly formed bone.Results At 4 weeks after surgery the osteosynthesis site was surrounded with more amounts of fibroconnective tissues in experimental group than that in control group.X ray observation showed that the medullary cavity of osteosynthesis site was connected,new bone was reconstructed at 12 weeks after surgery in experimental groups.New bone was greater in experimental group than that in the control group at 8,12 weeks after surgery(P<0.05).Conclusion Systemically administering rOGP may play some significant roles in the healing of bone allograft in Japan white rabbits.
