摘要 为探讨12b-羟基-去-D-杰氏山竹子素A(bHGA)的体外潜在成骨活性,用bHGA处理MC3T3-E1成骨细胞,研究了其对细胞碱性磷酸酶活性、矿化、骨相关基因表达及早期成骨分化相关基因表达水平的影响.结果表明:bHGA处理细胞后,对Runx2 mRNA表达无显著影响,而对Osx mRNA的表达具有一定抑制作用.bHGA能同时促进骨钙蛋白基因OCN和Ⅰ型胶原Col 1 mRNA的表达,但抑制碱性磷酸酶mRNA的表达,其对骨桥蛋白基因OPN mRNA的表达没有显著影响.说明bHGA不利于骨早期分化,但它通过促进骨钙蛋白和I型胶原mRNA的表达而促进骨成熟. To study the potential osteogenetic activity of 12b-hydroxy-des-D-garcigerr in A ( bHGA) in vitro, MC3T3-E1 was treated with bHGA and the alkaline phosphatase activity , mineralization, the expression of bone-related genes and the early osteogenic differentiation-related genes of MC3T3-E1 cells were measured.The results showed that the mRNA expression of Runx2 was generally not affected by bHGA , however, it inhibited the mRNA expression of Osx.bHGA promoted the expression of OCN and Col 1 but inhibited the mRNA expression of APL.OPN expression was not affected by bHGA.Our study demonstrated that bHGA was not conductive to the early differentitation of bone , while it might stimulate bone maturation by enhancing OCN and Col 1 mRNA expression .
