神经调节素-1β对大鼠脑局灶性缺血再灌注后白质损伤的保护作用

Protective effect of neuregulin-1β on cerebral white matter after cerebral focal ischemia reperfusion in rats

摘要 目的:研究神经调节素-1β(NRG-1β)对大鼠脑局灶性缺血再灌注脑白质损伤的保护作用.方法:SD大鼠随机分为假手术组,缺血再灌注组即MACO/A组和NRG-1β干预组.H-E染色观察脑白质组织病理学病变;透射电镜观察少突胶质细胞和髓鞘的改变.应用酶联免疫法(ELISA)检测脑白质肿瘤坏死因子-α(TNF-α)含量;免疫组织化学显色检测髓鞘碱性蛋白(MBP)的表达;原位切口末端标记技术(TUNEL)标记凋亡细胞的数目.结果:与假手术组相比,缺血再灌注组缺血侧脑白质囊性变,大量炎症细胞浸润,电镜下可见脱髓鞘改变.MBP阳性分布减少,TNF-α含量增高,凋亡细胞增多与假手术组相比差异均有统计学意义.NRG-1β干预组与缺血再灌注组相比,H-E染色显示缺血侧脑白质囊性变减少,炎症细胞浸润减少,电镜下脱髓鞘改变减少.MBP阳性分布有增多,TNF-α含量减少,凋亡细胞减少差异均有统计学意义.结论:大脑中动脉缺血再灌注可对脑白质造成显著损伤.侧脑室注射NRG-1β干预治疗对脑缺血再灌注后的脑白质具有保护作用,其机制与抑制炎症反应、减少胶质细胞凋亡和髓鞘破坏相关.
