摘要
为了解四川省副猪嗜血杆菌病的流行情况及分析分离株espp2基因,本试验从2013年8月到12月分离该菌并扩增其espp2基因进行序列分析。结果显示,分离到12株副猪嗜血杆菌,镜检发现为革兰阴性短杆菌;卫星试验表明分离株在金黄色葡萄球菌周围产生卫星现象;生化试验显示分离株发酵葡萄糖、麦芽糖、半乳糖、蔗糖、果糖、木糖、甘露醇、山梨醇、尿素、D-核糖和精氨酸水解酶;PCR鉴定发现均扩增到约821bp的目的片段。药敏试验显示分离株对氟苯尼考、先锋霉素V、阿莫西林、强力霉素和环丙氟哌酸敏感。扩增SC-1和12株分离株的espp2基因并测序,测序结果与GenBank公布的副猪嗜血杆菌SH0165、ZJ0906和Nagasaki的espp2基因共计16条序列进行比对发现相似性大于98%,这16条序列编码的氨基酸序列相似性大于97%,表明副猪嗜血杆菌espp2基因变异度低,较保守。
In order to find out prevalence of Haemophilusparasuisdisease in Sichuan province and analyze the espp2 gene of Haemophilus parasuis isolates,Finally 12 Haemophilus parasuis strains were isolated from August to December 2013,all the isolates were Gram-negative;fermented glucose,maltose,galactose,sucrose,fructose,xylose,mannitol,sorbitol,urea,D-ribose and arginine hydrolase;target fragment of 821 bp were amplified by PCR,the indicating that the isolates were Haemophilus parasuis.The isolates were sensitive to florfenicol,cephaloridine V,amoxicillin,doxycycline and ciprofloxacin.The espp2 genes of the 13 isolates including the SC-1,aHaemophilus parasuis strain isolated in 2009,were amplified and sequenced.The espp2 gene sequence of Haemophilus parasuis strains SH0165,ZJ0906 and Nagasaki shared 98% homology,and 16 amino acid sequences was larger than 97% similarity,indicating that the variability of the espp2 gene was low and the gene was conservative relatively.
