摘要
【目的】对新疆不同品种葡萄及葡萄干白藜芦醇含量进行HPLC测定,研究不同品种葡萄制干前后白藜芦醇的含量及变化规律。【方法】采用高效液相色谱法,色谱条件为Ultimate XB-C18色谱柱(4.6×250mm,5μm),流动相甲醇-水(45:55,V/V),流速0.8 mL/min,柱温30℃,检测波长306 nm。测定了40个品种葡萄及53个品种葡萄干的白藜芦醇含量。【结果】该方法测定白藜芦醇的平均保留时间为13.80 min,平均回收率为103.2%,最低检出限为0.8μg/mL。采用该方法测定的40个品种葡萄果实白藜芦醇平均含量为4.406μg/g,最高9.041μg/g,最低2.680μg/g。53个品种葡萄干白藜芦醇平均含量为3.569μg/g。最高6.295μg/g;最低1.594μg/g。【结论】该方法分析速度快,具有较高的重现性及灵敏度,适用于葡萄及葡萄干中白藜芦醇的测定分析。
【Objective】In order to determinate resveratrol content in different grape varieties by HPLC. 【Method】HPLC was performed on a Ultimate XB- C 18 column( 4. 6 × 250 mm,5μm),using methanol and water solution( 45: 55) as mobile phase at the flow rate of 0. 8 mL / min and the detection wavelength of 306 nm. 【Result】The method had the retention time of 13. 80 min,the average recovery was 103. 2%,and the limits of detection was 0. 8 μg / mL. The resveratrol average content was 4. 406 μg / g in 40 varieties of grapes by the method,the highest 9. 041 μg / g,and the minimum 2. 680 μg / g. The resveratrol average content was 3. 569 μg / g in 53 varieties of raisins,the highest 6. 295 μg / g,and the minimum 1. 594 μg / g. 【Conclusion】The method was fast,reproducible and practical for the routine analysis of resveratrol in grapes and raisins.
