新疆不同地区不同来源细粒棘球蚴原头节几种同工酶的等电聚焦分析

Isoelectro-Focusing Analysis of Several Isozymes of Protoscolices of Echinococcus granulosus from Different Sources in Xinjiang,China

摘要 本文报告用等电聚焦方法对新疆和田、乌鲁木齐和阿勒泰三地绵羊及伊犁和阿勒泰两地的黄牛细粒棘球蚴(Echinococcus granulosus)原头节标本进行苹果酸脱氢酶(MDH)、苹果酸酶(ME)、莆萄糖磷酸异构酶(GPI)、酯酶(EST)和酸性磷酸酶(ACP)同工酶谱的分析。实验结果表明:在新疆不同地区绵羊中传播的E.granulosus 存在着以和田与乌鲁木齐羊源为代表的两个在同工酶酶谱上有明显差异的虫株;和田羊源与阿勒泰羊源的同工酶谱十分相近;阿勒泰和伊犁两地牛源样本有显著不同,这表明至少在同一种中间宿主中可能也有生物学性状不同的变异株;阿勒泰羊源与牛源标本酶谱的相似性说明存在有同一虫株感染不同宿主的可能性. An analysis of isozymograms of enzymes EST、ME,GPI,MDH and ACP was made on protoscolices of sheep Echinococcus granulosus from Hotan,Urumqi and Altay of Xinjiang and of cattle E.granulosus from Ill and Altay,using isoelectrofocusing electrophoresis. Isozymograms of EST and ME for isolates from sheep of these three areas showed basic con- sistency,and isozymogram of MDH for Urumqi isolates from sheep was very different from that for Hotan and Altay isolates from the same source,the Sm values being 0.33 for both Urumqi isolates and Hotan and Altay isolates.There was also a difference in GPI isozymo- gram:the PI of dominant enzyme band was 5.55 for Urumqi isolates and 0.61 for Hotan and Altay isolates.For isolates from cattle,the dominant enzyme band of GPI was pH 5.55 for Ili isolates and pH 6.1 for Altay isolates.On EST isozymogram,the four highly active domi- nant enzyme bands of Ili isolates from cattle were pH 4.82,pH 5.34,pH 5.65 and pH 5.9, respectively;and the three main enzyme bands of Altay isolates from cattle were pH4.82, pH 5.9 and pH 6.2,respectively.Zymograms of isolates from sheep of different areas were very close:the Sm value of Hotan and Altay isolates was 0.957 and that of Urumqi isolates was 0.597;but zymograms of isolates of cattle origin were very different:the Sm value of Altay and Ili isolates was 0.505.Zymograms of isolates from different sources of the same area were more similar,findings of Altay isolates from sheep and from cattle being similar (Sin=0.692).Zymograms of isolates from different sources of different areas showed great difference:Urumqi isolates from sheep and Altay and Ili isolates from cattle displayed dis- tinct difference,Sm values being 0.529 and 0.420,respectively,and Hotan isolates from sheep and Ili isolates from cattle had an Sm value of 0.523.One exception was that zymo- grams of Hotan isolates from sheep and Altay isolates from sheep and cattle gave similar re- sults,Sm values being 0.957 and 0.650,respectively.No differences in ACP isozymograms were fot,nd among
