摘要
目的 采用流式细胞仪检测复方北芪口服液对大鼠大脑中动脉阻塞 /缺血再灌流(MCAO/IR)模型细胞凋亡的影响 ,从细胞凋亡的角度探讨该药作用于脑缺血损伤的可能机制。方法 清洁级雄性SD大鼠 18只 ,随机分为模型组、假手术组、治疗组。取新鲜缺血侧脑海马制成单细胞悬液 ,进行凋亡的定量分析。结果 假手术组即可测得凋亡峰 ,模型组、治疗组细胞凋亡百分率明显高于假手术组 ,均有显著性差异 (P <0 .0 1) ;治疗组的百分率较模型组明显降低 ,有显著性差异 (P <0 .0 1)。结论 复方北芪口服液有明显抗凋亡的作用 。
Objective It is to measure the effect of Compound Beiqi Mixture (CBM) on apoptosis in rats with middle cerebral artery occlusion and ischemia reperfusion (MCAO/IR) with flow cytometry (FCM), and to discuss the possible mechanism of CBM on ischemia injury from the direction of apoptosis.Methods 18 male rats were divided into model group, sham-operated group and treatment group. Hippocampus in ischemic side was chosen and made into unicellular suspension. Quantitative analysis for apoptosis was progressed.Results Apoptosis peak was detected in the sham-operated group. The apoptosis percentages in the model group and treatment group were obviously higher than that in the sham-operated group, and there were significant difference (P<0.01). The apoptosis percentages in the treatment group was obviously lower than that in the model group, and there was significant difference (P<0.01).Conclusion CBM has significant anti-apoptosis effect. It has prevalent directive meaning that ischemia apoplexy is treated with Bushenyiqihuoxue method.
