摘要
目的 观察 β葡聚糖是否可以促进抗体产生并探讨其作用是否依赖补体。 方法 用 β葡聚糖与抗原OVA联合免疫C5 7BL 6小鼠和补体C3基因缺陷小鼠 ,ELISA实验检测总IgG、IgG2a、IgG1抗体的产生。结果 β葡聚糖与OVA联合免疫的C5 7BL 6小鼠 ,其OVA特异性IgG、IgG2a、IgG1抗体应答均显著高于单独用OVA免疫的小鼠 ,低于完全弗氏佐剂与OVA免疫的小鼠 ;补体C3基因缺陷小鼠IgG、IgG2a、IgG1抗体滴度均显著低于野生型C5 7BL 6小鼠。结论 β葡聚糖可以促进抗体产生 ;补体C3缺损显著影响 β葡聚糖介导的抗体应答。
Objective To observe whether β glucan can enhance the production of antibody and explore the roles of complement system in this effect. Methods C57BL/6 and C3 deficient mice were immunized with β glucan and antigen OVA complexes. The production of IgG, IgG2a, and IgG1 was determined by ELISA. Results OVA specific IgG, IgG2a, and IgG1 antibody responses in mice immunized with β glucan and antigen OVA were significantly increased than those in the mice immunized with OVA alone, and were slightly lower than those in mice immunized with an emulsion of complete Freund's adjuvant containing OVA. The titers of IgG, IgG2a, and IgG1 in C3 deficient mice were significantly lower than those in wild type mice. Conclusion β glucan can enhance the antigen response and deficiency of complement C3 can significantly impair the antigen response mediated by β glucan.
