摘要
目的:从国产蝮蛇毒中分离提取蛋白C活化组份。方法:以江浙蝮蛇毒为材料,经DEAE-SephadexA-50、CM-SephadexG-25二次柱层析,分离提取蛋白组份,并用KlPTT法及发包庇物法检测其抗凝活性及酰胺酶活性。结果:经二次柱层析,分离出了FⅥ-Ⅲ蛋白组份,经生物学活性鉴定表明,该组份具有强烈的抗凝血活性及酰胺酶活性,同时观察不同浓度FⅥ-Ⅲ组份对正常血浆KFPTT值及A405值的影响,发现KPTT值随FⅥ-Ⅲ组份浓度的增加而升高,两者呈正相关,A405值也与该组份有显著量效关系,表明我们分离提取的蛇毒组份FⅥ-Ⅲ具有抗凝活性,并且该活性与蛋白C的活化有关。
To isolate and chalacterize a fraction with protein Gactivating activity from Agkistrodon haly venom (from Jiangshu Province, China). Metheds: Through successiveionexchange column chIOInatgraphy of DEAE-Sephadex A-50 and of CM-Sephadex C-25, aprotein-containing fraction was isolated from Agkistrodon haly venom, and its possibleanticoagulant and dridase activities were measured by methed of KPTT and that of specificchromogenic substrate, respectively. Results: After the two-time column chromatography, fractionFⅥ-Ⅲ Which was protein-containing was isolated, and the identification of its biologicalactivities resulted in the finding of potent anticoagulant and dridase activities. Moreover, theeffects of different concentrations Of fraction FⅥ-Ⅲ on nounal human plasma KPTT values andthe absorbance on 405urn (A405) were also studied, the results showed that the KPTT valuesincreased along with the elevation of fraction FⅥ-Ⅲ concentrations, appleing peitivelycorelative to each other, and the A405 also shiwed sighficantly concentration-dependent. Theabove results indicate that the isolated fraction possesses anticoaedant activity, which isrelative to the activation of protein C.
