摘要
目的:观察冠通胶囊对小鼠减压缺氧,大、小鼠脑缺血时血中儿茶酚胺水平的影响。方法:健康 ICR 小鼠在水合氯醛麻醉下,于真空瓶中减压10 min 造成缺氧或 SD 大鼠、ICR 小鼠分别结扎双侧带迷走神经的颈总动脉20、8 min 造成脑缺血,用高效液相色谱-电化学联合检测法测定缺氧、缺血动物血浆中儿茶酚胺水平。结果:缺氧、缺血小鼠血浆中去甲肾上腺素和肾上腺素浓度显著增加(P<0.01),灌胃冠通胶囊1h 后缺氧小鼠血浆肾上腺素水平、缺血小鼠血浆肾上腺素和去甲肾上腺素水平显著下降,大鼠脑缺血20 min仅见肾上腺素水平增加(P<0.05)。结论:小鼠缺氧或脑缺血可使交感神经激活导致血中去甲肾上腺素和肾上腺素释放增加,冠通胶囊可拮抗之。
Objective:To investigate the effect of Guantong Capsule on plasma catecholamine levels in mice during anoxia and cerebral is- chemia in mice and rats.Methods:Experiments were conducted in 20-22g ICR mice under chloral hydrate anesthesia and placed the mice in vacuum bottle with 0.07 mPa for 10 min to cause anoxia.Cerebral ischemia models of SD rats and ICR mice induced by bilater- al common carotid ligation for 20 min and 8 min respectively.HPLC combined with electrochemical assay was used to determine cate- cholamine level in anoxic and ischemic plasma of model animal.Results:The norepinephrine (NE) and epinephrine (E) concentration in anoxic and ischemic mice plasma obviously increased (P<0.01),1 h after orally taken Guantong Capsule,the plasma level of NE in anoxic mice,and plasma E and NE in ischemic mice were all recovered to normal control level (P<0.05),only the E level elevation was observed in cerebral ischemic rats for 20 min (P<0.05).Conclusion:The anoxia or cerebral ischemia in mice could activate the sympathetic nerve which induce the increase of plasma NE and E release,the Guantong Capsule could antagonize it.
