摘要
在成功构建斑马鱼肌球蛋白轻链2启动子转红色荧光蛋白基因唐鱼Tanichthys albonubes可遗传品系的基础上,作者采用荧光显微镜观察和用RT-PCR技术对外源基因在唐鱼体内的表达情况进行检测。结果表明:红色荧光蛋白基因在F3代唐鱼体内的表达最早出现在受精后19 d,多数个体的表达集中在25~30日龄,最晚的表达个体出现在35日龄;红色荧光蛋白的表达主要分布于肌肉、心脏、肝脏、脾、鳃、鳔、肠和性腺,在肌肉、肝脏、鳃组织中的相对表达量较其它组织高,而在表皮和鳍条中没有检测到外源基因的表达。虽然红色荧光蛋白在转基因唐鱼后代出现表达迟缓、表达特异性改变的现象,但在外源基因后代中的遗传和表达在总体上还是稳定的,有望培育出转红色荧光蛋白基因唐鱼品系。
Fluorescence microscopy and RT-PCR technique were used to detect the expression of exogenous gene in the genetic strains of white cloud mountain minnow Tanichthys albonubes successfully constructed and modified by recombinant plasmid pDsRed-mylz2 from the zebrafish inserted myosin light chain 2 promoter and red fluorescent protein gene.The results showed that the expression of the red fluorescent gene was initially observed 19 days after fertilization in F3,and expressed mostly in the 25-30 days,the latest expression of the genetically modified fish appeared in the 35 days.The red fluorescent protein was expressed in muscle,heart,hepatopancrease,spleen,gill,swim bladder,intestines and gonad,and the expression was much stronger in the muscles,hepatopancrease,and gill than that in other tissues,but the fluorescence was not detected in epidermis and fin ray.Although postponement of the red fluorescent protein expression and the change in specificity was appeared occasionally in transgenic offspring of the minnow,the genetic expression of the exogenous gene in the offspring was stable in general.So it is expected to produce a stable strain of red fluorescent protein transgenic white cloud mountain minnow.
