摘要
目的重组chemerin蛋白对3T3-L1脂肪细胞visfatin、MCP-1等基因表达的影响。方法 3T3-Ll前脂肪细胞诱导分化至第10天成熟后,分别以不同浓度(0、25、50、100、200 ng/ml)的chemerin蛋白干预12、24、48 h后收集细胞,RT-PCR法检测visfatin、MCP-1等基因的表达情况。结果 chemerin蛋白作用于成熟的3T3-Ll脂肪细胞24 h后,100 ng/ml干预组visfatin的表达与对照组比较差异有统计学意义(P<0.05),50、100、200 ng/ml的chemerin蛋白干预组MCP-1表达与未干预组比较差异显著(均P<0.05);作用48 h后,50、100、200 ng/ml的chemerin蛋白干预组与对照组相比均上调了visfatin与MCP-1的表达(均P<0.05)。结论重组chemerin蛋白上调成熟的3T3-Ll脂肪细胞visfatin与MCP-1基因的表达。
Objective To investigate the effects of recombinant chemerin protein on visfatin and MCP-1 expression in 3T3-L1 adipocyte.Methods After 3T3-L1 preadipocytes were differentiated 10 days,mature adipocytes were treated with different concentration of chemerin(0,25,50,100,200 ng/ml) at various time periods(0,12,24,48 h).The expressions of visfatin and MCP-1 were detected by reverse transcription polymerase chain reaction(RT-PCR).Results Compared with that of control group,100 ng/ml chemerin increased visfation expression after 24 h treatment,at the same time,50,100,200 ng/ml chemerin also increased MCP-1 expression(P<0.05).The expressions of visfation and MCP-1 were significantly higher at the dose of 50,100,200 ng/ml of chemerin(P<0.05) after 48 h treatment.Conclusions Recombinant chemerin protein could increase the expression of visfatin and MCP-1 in mature 3T3-L1 adiopocytes.
