To study the antitumor effect of biodegradable 5-fluorouracil-loaded polylactic acid-nanoparticales (5-Fu-PLA-NP) on human gastric and colorectal carcinoma cell lines.Methods 5-Fu-PLA-NP were prepared with the method of ultrasound emulsification.By MTT assay,the cytotoxic effects of 1×10 -7 mol/L,1×10 -6 mol/L,1×10 -5 mol/L,1×10 -4 mol/L 5-Fu-PLA-NP and 5-Fu on human gastric cell line MGC803 and human colorectal carcinoma cell line SW620 were detected in vitro on 10 consecutive days.The 50% inhibition concentrations values (IC50) and inhibitory rate of these two drugs against the MGC803 and SW620 cells were calculated.Results The cumulative release rates of 5-Fu-PLA-NP were 75.7% at 7th day and 94.3% at 10th day in vitro.The cytotoxic effect of 5-Fu showed a time-dependent manner from the first day to 7th day,and then reached a plateau.But cytotoxic effect of 5-Fu-PLA-NP sustained this manner until the 10th day.Both 5-Fu-PLA-NP and 5-Fu showed dose-dependent cytotoxic effects at the 7th day,and the effects of these two drugs were not obviously different.Conclusion The 5-Fu-PLA-NP has drug sustained-release character and can prolong the phase of the inhibitory effect of 5-Fu against human gastric and colorectal carcinoma cells.The 5-Fu-loaded nanoparticles do not reduce the biological activity of 5-Fu.4 refs,2 figs.
