大孔吸附树脂分离纯化冬凌草甲素条件的优化 被引量:2

摘要 比较了大孔吸附树脂D-101、DM-130、AB-8和NKA-9对冬凌草甲素的吸附和解吸附能力。结果表明,AB-8树脂对冬凌草甲素具有较好的吸附分离性能。其最佳分离纯化工艺条件为,吸附时进料浓度0.486 mg/mL、流速2 BV/h、进样量4 BV;解吸附时体积分数80%的乙醇溶液作洗脱剂、流速1.5 BV/h、洗脱剂用量3 BV。经AB-8树脂吸附与解吸附后,冬凌草甲素回收率为78.4%;用甲醇重结晶后纯度达到99.5%,回收率为54.3%。 The adsorption and desorption capacities of four kinds of macroporous adsorption resins,D-101,DM-130,AB-8 and NKA-9 were compared.The results showed that AB-8 resin had the best adsorption and desorption capability.And the optimum technology parameters for adsorption were feeding concentration 0.486 mg / mL,flow rate 2 BV / h and feeding volume 4 BV;for desorption were 80%(volume fraction) alcohol as elution solvent,flow rate 1.5 BV / h and elution volume 3 BV.After treated with AB-8,the yield of rubescensine A was 78.4%.And the purity of rubescensine A could be up to 99.5% if recrystallized by methanol,and the yield was 54.3%.
