摘要
目的:探讨曲克芦丁原料药对TA97、TA98、TA100、TA102和TA1535菌株基因回变菌落的影响,对其遗传安全性进行评价。方法:采用直接平皿掺入法,将曲克芦丁原料药以5、1、0.2、0.04、0.008mg曲克芦丁/皿为终浓度,在活化与非活化条件下,分别与鼠伤寒沙门氏菌组氨酸缺陷型突变菌株,37℃接触48小时,计数回变菌落数。结果:在5~0.008mg曲克芦丁原料药/皿的不同测试浓度,+S9与-S9两种测试系统条件下,其作用的5种测试菌株的回变菌落数与阴性对照相比无明显增加,回变菌落背景正常,未显示其有抑菌作用,且无剂量-反应关系。结论:曲克芦丁原料药在本试验所确定的5mg/皿剂量范围内,对TA系列测试菌株无明显致DNA碱基置换及移码突变性。
Objective:To investigate the effect of Troxerutin on bacterial strains TA97,TA98,TA100,TA102 and TA1535 of genie mutation and study hereditary toxicity of them.Methods:The bacterial strains TA97,TA98,TA100,TA102 and TA1535 were respectively contacted with 0.008 to 5 mg/a utensil final concentration doses of Troxerutin under mixing S9 or non-mixing S9 and 37℃ exposure to 48 hours,the DNA variety of TA bacterial was observed.Results:The TA bacterial reverse mutation and induced by Troxerutin was not increased under mixing S9 or non-mixing S9,compared with that in control group.Conclusion:The Troxerutin in the range of 5 mg/a utensil final concentration were not genotoxicity to TA bacterial aberration.
