摘要
在一定的温度、渗透压、离子强度条件下,利用脂质体材料L-α-phosphatidylcholine制备人工脂质体,借助结晶紫染液、美蓝染液、复红染液、钼蓝染液、苏丹黑染液,对脂质体进行染色,结果表明经结晶紫染色后的脂质体在光学显微镜下具有形态大小分明、数量多、立体结构清晰等特征.为了优化染色方案,进一步获取最佳染色配比,实验进行了梯度染色,得到了背景清晰、脂质体立体结构明显的最佳染色方法,解决了光学显微镜下不易分辨脂质体的问题,为后续实验的开展奠定基础.
Artificial lipsomes was performed under appropriate conditions such as temperature,osmotic pressure,ionic intensity.The results showed that artificial lipsomes which stained by crystal violet staining solution characterised as legible shapes and stereochemical structures,numerous quantity under optimal microscope with dyeing of crystal violet staining solution,methylene blue solution,basic fuchsin solution,molybdenum blue solution,sudan black solution.The gradient dyeing was investigated,demonstrating that proper proportion was optimum resulting from optimizing the staining model.The research made a solution to distinguish artificial lipsomes on the optimal microscope,which was in favour of subsequent experiment.
