摘要
目的探讨阴虚动风证帕金森病LID大鼠脑内氨基酸类神经递质的动态变化及复方地黄方的干预作用。方法采用6-羟基多巴胺(6-OHDA)偏侧损毁黑质制备帕金森病(PD)大鼠模型,成功的PD模型大鼠腹腔注射左旋多巴/苄丝阱[50/12.5(mg.kg)],制备阴虚动风证帕金森病LID病证的大鼠模型。将大鼠分为复方地黄方组、LID组、正常对照组、假手术组,并在治疗4周、6周后取纹状体,应用高效液相法(HPLC)测定各组大鼠纹状体内氨基酸的含量。结果阴虚动风证帕金森病LID模型大鼠纹状体内兴奋性氨基酸显著升高;复方地黄方能显著降低兴奋性氨基酸的含量,抑制性氨基酸含量则明显升高。结论复方地黄方缓解帕金森病LID的机制可能是通过调节脑内氨基酸的水平、减轻其兴奋毒性来实现的。
Objective To study the dynamic changes and effects of the compound rehmannia formula on the content of amino acid neurotransmitters in the brain of the lovodopa-induced dyskinesia(LID) model of Yin-deficiency and Wind-agitation.Methods The Parkinson's disease(PD) rat models were established through stereotoxic injection of 6-hydroxydopamine.The successful PD models were injected with levodopa/beserazide[50/12.5(mg·kg)] to establish the LID model of Yin-deficiency and Wind-agitation.The rats were randomly divided into LID model group,the compound rehmannia formula group,normal control group and sham-operation group.Amino acid content in the rat striatum was determined with HPLC after treatment for 4 and 6 weeks,respectively.Results The content of EAA of LID was significantly increased in Yin-deficiency and Wind-agitation model group.In the compound rehmannia formula group,the content of EAA was significantly decreased but the content of IAA was obviously increased.Conclusion The compound rehmannia formula can alleviate LID in Parkinson's disease probably by adjusting the amino acid content in the brain and mitigating the excitotoxicity.
