Expression and clinical significance of matrix metalloproteinase-9 in acute hepatic failure

摘要 目的探讨基质金属蛋白酶-9(MMP-9)在急性肝炎肝衰竭中的表达及其临床意义。方法 2009~2011年共收集急性肝炎肝衰竭患者的肝组织标本17例、正常肝组织标本21例,通过RT-PCR检测MMP-9的mRNA表达;用明胶酶谱法检测肝内MMP-9和pro-MMP-9的活性变化;West-ernblot测定其蛋白表达量的变化。结果急性肝炎肝功能衰竭患者肝脏中MMP-9的mRNA水平、蛋白水平和酶的活性明显高于正常人。结论 MMP-9在急性肝炎肝功能衰竭的肝脏组织标本中表达显著增高,活性也明显高于正常,其有可能成为急性肝损伤时的潜在治疗靶点。 Objective To investigate the expression and clinical significance of matrix metalloproteinase-9(MMP-9)in acute hepatic failure.Methods Specimens of acute hepatic failure were obtained freshly from patients undergoing liver transplantation with approval from ethics committee in our institution.The normal liver tissue was obtained from the donor.The methods of RT-PCR and Western blot were used to analyze the expression level of MMP-9 mRNA and protein in acute liver failure group and normal group respectively.The activity of MMP-9 and pro-MMP-9 in hepatic tissue were assayed by zymography.Results The expression levels of MMP-9 mRNA in the acute liver failure were significantly higher than those in normal liver(P<0.05).The zymography and western blot showed similar changes on that of the mRNA.Conclusions MMP-9 is up-regulated in acute hepatic failure patients.MMP-9 may play an important role in the acute hepatic failure.
