摘要
【目的】探索V形孔培养法(WOW)、改进V形孔培养法(mWOW)和常规微滴培养法对牛胚胎体外生产效果的影响。【方法】将牛卵母细胞和受精卵分别采用常规微滴培养法(对照组)、WOW和mWOW进行体外培养,对照组每个微滴中置入20~25枚卵母细胞或胚胎,WOW和mWOW组每个V形孔中置入1枚卵母细胞或胚胎,计算卵裂率、囊胚率及受精后第6,7,8天的囊胚总细胞数和囊胚孵化率。【结果】mWOW组的卵裂率、囊胚率及第6,7,8天囊胚的总细胞数、囊胚孵化率与常规微滴组差异不显著(P>0.05),但显著高于WOW组(P<0.05);3个组第6,7,8天获得的囊胚总细胞数均呈逐渐下降的趋势。【结论】以TCM-199为基础培养液,用mWOW可以完成牛胚胎体外生产的全过程,且比WOW培养胚胎具有更强的发育能力和更好的质量。体外培养时,发育越快的胚胎质量越高。
