摘要
目的 探讨丙型肝炎病毒(HCV)核心蛋白对肝门部胆管癌生长和转移的影响。方法 首先构建表达HCV核心蛋白的真核表达载体pcDNA3-HCVcore,再将含有 pcDNA3-HCVcore的载体和不含 pcDNA3-HCV core的空载体分别导入肝门部胆管癌细胞株 QBC939中,RT-PCR和免疫组织化学检测 VEGF mRNA和蛋白表达。结果 重组质粒 pcDNA3-HCVcore在 QBC939细胞中有稳定表达;表达 HCV核心蛋白的细胞QBC939的 VEGF在蛋白水平和 mRNA水平较转染空载体的细胞QBC939明显升高。结论 HCV核心蛋白能激活VEGF表达,可能对肝门部胆管癌的生长和转移具有促进作用。
Objective To study the effect of HCV core protein on the growth and metastasis of hilar cholangiocarcinoma. Methods The eukaryon expression vector pcDNA3-HCV core, which could express HCV core protein, was firstly Constructed. The vector with pcDNA3 - HCV core and the vector without pcDNA3-HCV core were transducted into hilar cholangiocarcinoma celi strain (QBC 939) separately. VEGF mRNA and VEGF protein were detected by RT-PCR and immunohistochemistry respectively. Results The recombinant plasmids with pcDNA3-HCV core were stably expressed in QBC 939 celis. The VEGF mRNA and VEGF protein expressed in those QBC 939 celis transducted with pcDNA3-HCV core were higher than that of QBC 939 celis without pcD-NA3-HCV core. Conclusion HCV core protein can activate the expression of VEGF in QBC 939 celis and might contribute to the growth and metastasis of hilar cholangiocarcinoma.
