摘要
目的探索磷脂酰肌醇3激酶(PI3K)抑制剂雷帕霉素对胶质瘤干细胞的特异性凋亡作用。方法应用磁式分选系统将胶质瘤细胞分为CD133+和CD133-细胞。应用MTT法检测雷帕霉素对两种胶质瘤细胞的生长抑制作用。Annex-in V-FITC和碘化丙锭(PI)双染检测细胞凋亡。流式细胞仪检测细胞周期。Western blot检测PI3K蛋白表达。结果雷帕霉素以浓度依赖方式抑制胶质瘤细胞的生长,且对CD133+细胞的生长抑制作用显著强于CD133-细胞。比起CD133-细胞,雷帕霉素对CD133+细胞具有更强的凋亡作用。结论雷帕霉素对胶质瘤干细胞具有特异性的抗肿瘤效应,可能与胶质瘤干细胞具有较强的PI3K活性有关。
Objective To explore glioma stem cell-specific apoptosis induced by PI3K inhibitor rapamycin. Methods Magneticseparation system was applied to separate glioma cells into CD133 positive and CD133 negative cells. MTT assay was applied to test growth inhibition of CD133 positive and CD133 negative cells by rapamycin. Annexin V-FITC and propidium iodide (PI) double staining was used to detect apoptosis. Cell cycle was detected by flow cytometry. PI3K protein expression was detected by Western blot. Results Rapamycin can inhibit the growth of glioma cells in a dose-dependent manner. The growth inhibition of CD133 + cells was significantly stronger than that of CD133 -cells. Compared to CD133 -cells,CD133 + cells had a stronger apoptosis rate induced by rapamycin. Conclusions Rapamycin have specific anti-tumor effect on glioma stem cells,which may be related to strong PI3K activity in glioma stem cells.
