摘要
目的观察复方感冒合剂对孕鼠及胚胎发育毒性的影响。方法将50只SD孕鼠随机分为5组,即复方感冒合剂高(13.0g/kg)、中(6.5g/kg)、低(3.3g/kg)3个剂量组、阴性对照组和阳性对照组,每组10只,在大鼠受孕的第7-16天分别灌胃给予复方感冒合剂、生理盐水和新康泰克,所有孕鼠在妊娠的第20天处死,剖腹取子宫称重,记录总胎数、吸收胎数、死胎数和活胎数;测量并记录活胎鼠体重、身长和尾长,并检查胎鼠外观有无异常;每窝50%的活胎鼠经茜素红和阿利新蓝染色后做骨骼检查,50%用Bou in液固定后做内脏检查。结果复方感冒合剂各剂量组孕鼠的体重增加、生殖能力,胎鼠的体重、身长和尾长等生理指标的变化与阴性对照组比较差异无统计学意义(P>0.05);且各剂量组与阴性对照组比较,胎鼠的外规、骨骼、内脏发育均未见明显畸形,差异无统计学意义(P>0.05)。结论在本实验室条件下,复方感冒合剂各剂量均无明显母体毒性、胚胎毒性和致畸性,且在实验剂量范围内,复方感冒合剂未呈现剂量反应关系。
Objective To observe the toxic influences of Fufang Ganmao Mixture(Compound Mixture for Common Cold) on pregnant rats and embryonic development.Method 50 SD pregnant rats were randomly divided into 5 groups: 3 Fufang Ganmao Mixture groups(high dose,at 13.0g/kg;medium dose,at 6.5g/kg and low dose,at 3.3g/kg),negative control group and positive control group.Each group was made up of 10 rats,which were given Fufang Ganmao Mixture,normal saline and CONTAC NT(compound pseudoephedrine hydrochloride sustained-release capsules) via gastric perfusion at 7 to 16 days after pregnancy.Then,all pregnant rats were killed at the 20th day.The uterine weight,numbers of total fetus,absorbed fetus,dead fetus and fetus alive were recoded.The rat body weight,body length and tail length were measured and recorded,with examination for abnormality of fetus appearance.50% of fetus alive in each cage underwent skeletal examination via stained by alizarin red and alcian blue,the other 50% were fixed with Bouin and underwent visceral examination.Result No statistical difference existed in comparison between Fufang Ganmao Groups(all dose subgroups) and the negative control group at physiological indexes of increased body weight,reproductive capacity of the pregnant rats,as well as the fetus body weight,body length,tail length(P>0.05).And the appearance,skeletal and visceral development were observed no abnormality in rats of all dose Fufang Ganmao Groups,with no statistical difference compared with negative control group(P>0.05).Conclusion Under the condition of our lab,the Fufang Ganmao Mixture had no significant maternal toxicity,embryo toxicity and teratogenicity.In the experimental dose range,the mixture had no dose-related relationship.
