摘要
XY精子流式细胞分选技术是目前最有效的奶牛性别优化控制繁殖手段,其主要原理是根据奶牛携带Y染色体的精子和X染色体的精子,其DNA含量不同,通过荧光染料与DNA定量结合,用流式细胞技术将携带Y染色体的精子和携带X染色体的精子分开,实现X精子与Y精子的分选分离收集的效果,然后用于人工授精和生物胚胎制作。该技术已在欧美等国家使用,准确率达90%以上.我们在中国开发了该技术,结果优于国外。
XY sperm sorting in flow cytometry is currently the only effective method to select the sex of offspring in diary cow. This method is based on the difference of DNA content of X- and Y- chromosome-bearing sperm.The sperm sample is treated with a DNA-binding fluorochrome.Hoechst 33342 and sorted in flow cytometer into separate X and Y population, and then can be used for artificial insemination and embryo preparation.This technology has been used in several nations in the world and achieve over 90% of accuracy of the desired sex of offspring. We have developed this technology in China and showed better improvement than others.
