一种替代家兔热原试验的新方法初探——检测人全血中的TNF-α 被引量:3

摘要 为了寻找一种替代家兔热原试验的方法 ,最近我们对用人全血检测热原的可行性进行了初步探索。其原理是 :将细菌内毒素标准品与从健康自愿者提供的少量肝素抗凝全血共同孵育 6h后 ,用ELISA双抗夹心法测定其产生的内源性热原 (TNF -α)。试验结果表明 :TNF -α的量与内毒素的量在一定范围 (0 0 6EU mL~ 2 0 0EU mL)内有很好的相关性 ,不同来源的内毒素试验结果也是如此。说明该方法作为替代家兔热原检查的新方法有一定的可行性。 The aim of this study was to develop an in vitro test for the detection of pyrogen in order to replace the rabbit pyrogen test. Recently the human whole blood was assessed for tumor necross factor (TNF) α for the probability of the novel test. When human whole blood is incubated in the presence of reference standard endotoxin, they secrete “endogenous pyrogen' TNF α determined by ELISA after a fixed incubation time (6 hours). These results show that the correlation coefficient is greater than or equal to 0.980 in the range of 0 32EU/ml-200EU/ml between the TNF α and the endotoxin. The same result has got from the other kind endotoxin with equal effect in the LAL/TAL test. Therefore we think this human whole blood pyrogen assay will serve as aternative pyrogen test.
