摘要
目的 观察软骨细胞-新型高孔隙率聚乳酸( PLA)支架复合物在体内形成新生软骨的能力。方法 体外培养兔耳廓弹性软骨细胞,取第 2代软骨细胞与新型高孔隙率聚乳酸支架形成复合物,体外培养 1周后移植于兔颈部气管旁带状肌间隙,以未种软骨细胞的聚乳酸支架作为对照。结果 移植于兔体内 1个月后,接种了软骨细胞的新型高孔隙率 PLA支架有新生的软骨形成, 2个月后新生软骨逐渐成熟并形成明显的软骨陷窝,而对照组无软骨形成。结论 这种新型聚乳酸支架种植软骨细胞后在体内具有成软骨能力,可用作软骨组织工程的支架材料。
objective To observe the neocartilage formation in autograft chondrocytes- newstyle PLA polymer with higher porosity in vivo. Method The rabbit ear elastic chondrocytes were cultured in vitro and their 2nd generation chondrocytes were harvested and seeded in the new- style PLA polymers with higher porosity.After 1 week cultured,the autografts were transplanted into the muscles near the trachea of the rabbit and the PLA polymers without chondrocytes were cultured and implanted in the same way as controls.Result The higher porosity PLA polymers with autograft chondrocytes formed neocartilage 1 month After implantation and the neocartilage were mature after 2 months and bone lacuna was also formed,but there was no cartilage formation in the control.Conclusion The newstyle PLA polymer with higher porosity was suitable for the scaffads materials in the cartilage tissue- engineering.
