摘要
目的研究sonic hedgehog(Shh)信号通路各蛋白在血管平滑肌细胞(VSMC)中的表达情况,探讨其是否与VSMC的增殖相关。方法应用免疫荧光、激光共聚焦检测Shh信号通路蛋白在体外培养的VSMC中的表达,MTT法及Ki67的表达情况联合评价细胞增殖。结果Shh信号通路的配体Shh蛋白、patched1受体及其下游转录因子Gli2在VSMC中有表达,MTT法及Ki67染色结果显示外源的Shh蛋白能够促进VSMC的增殖,而应用Shh信号通路的特异抑制剂Cyclopamine则能抑制细胞增殖。结论Shh信号通路与VSMC增殖密切相关。
