摘要 目的研究sonic hedgehog(Shh)信号通路各蛋白在血管平滑肌细胞(VSMC)中的表达情况,探讨其是否与VSMC的增殖相关。方法应用免疫荧光、激光共聚焦检测Shh信号通路蛋白在体外培养的VSMC中的表达,MTT法及Ki67的表达情况联合评价细胞增殖。结果Shh信号通路的配体Shh蛋白、patched1受体及其下游转录因子Gli2在VSMC中有表达,MTT法及Ki67染色结果显示外源的Shh蛋白能够促进VSMC的增殖,而应用Shh信号通路的特异抑制剂Cyclopamine则能抑制细胞增殖。结论Shh信号通路与VSMC增殖密切相关。 Objective To study the expression of sonic hedgehog(Shh)signaling pathway in vascular smooth muscle cell(VSMC)and its effect on the proliferation of VSMC.Methods The expression of Shh signaling pathway in VSMC was detected by using immunofluorescence assay and laser confocal microscopy.The proliferation of VSMC was evaluted by combined application of MTT and Ki67 expression.Results The expressions of Shh protein,Patched-1 receptor,and transcription factor Gli2 were found in VSMC.Exogenous Shh protein promot...
