摘要 目的探讨IL-1(白细胞介素-1),TNF-α(肿瘤坏死因子-α)在牙根吸收中的作用是否等同于介导牙周炎性骨吸收的作用。方法建立机械损伤诱导牙根吸收大鼠模型,腹腔注射IL-1可溶性受体和TNF-α可溶性受体,应用组织形态技术观察抑制根吸收的病理形态影响,从逆反方向证实IL-1、NNF-α在诱导牙根吸收中的作用。结果 TNF-α组与病理对照组的牙根吸收陷窝数量、吸收陷窝长度、陷窝面积、位于根面牙周韧带、牙槽骨的TRAP(指多核破骨细胞,多核破牙骨质细胞,单核细胞,类成纤维细胞)阳性细胞计数各项指标均有显著性差异P<0.05,P<0.01)。而IL-1组的各项指标没有显著性差异(P>0.05)。结论 TNF-α是影响牙根吸收的主要细胞因子。 Objective In this study, the role of the in. ammatory cytokines interleukin - 1 (IL- 1) and tumor necrosis factor alpha (TNF- α) in the course of mechanically induced root resorption was investigated. Methods Mechanical induction of root resorption was performed on the upper left.rst molars in 18 male Wistar rats according to the method of Nakane and Kameyama. Starting on day minus 1, six animals received daily intraperitoneal injections of 2ml of 1 μg/ml soluble receptors to IL- 1 (sIL- 1RII) and another six animals were administered the same dose of soluble receptors to TNF - α (sTNFa - RI). Six animals served as a control. On the 7th day the left maxillae were prepared for histological and morphometric analysis of the extent of the root resorption that had developed. Results The qualitative and quantitative results demonstrated that in both receptor groups the amount of root resorption was significantly reduced. Especially following systemic application of sTNFa -RI, root resorption was nearly completely prevented. Conclusions Our results indicate that IL - 1 and more particularly TNF - α are important for the induction and the further process of mechanically induced root resorption in the rat.
