摘要
目的研究过氧化物酶增殖物激活受体γ(PPARγ)对肝癌细胞迁移的影响,并探讨其机制。方法应用伤口愈合实验观察PPARγ的合成配体曲格列酮和罗格列酮对肝癌细胞系BEL-7402迁移的影响;应用免疫荧光化学、RT-PCR和Western Blotting分析探讨其中的机制。结果曲格列酮和罗格列酮均能抑制肝癌细胞的迁移活动,其原因是由于该两种药物激活PPARγ通路且具有配体依赖性。结论PPARγ活化能够抑制肝癌细胞的迁移。
Objective To investigate the effect of peroxisome proliferators-activated receptor γ (PPARγ) on migration of hepatocellular carcinoma cells and its underlying mechanisms. Methods Migration of HCC cell line BEL-7402 with the treatment of troglitazone and rosiglitazone was measured by a wound healing assay. PPARγ mRNA and protein were assessed by RT-PCR, immunofluorescence chemistry, and Western Blotting analysis, respectively. Results Troglitazone and rosiglitazone inhibited the migration of BEL-7402 cells,both medcines activated PPARγ signaling pathway on ligand-dependent effect. Conclusion PPARγ activation inhibited the migration of HCC.
