摘要
目的:检测卵巢癌组织中EphA2受体mRNA及蛋白的表达水平。方法:应用激光捕获显微切割技术(LCM)辅助的一步RT-PCR及Western blot方法检测30例卵巢癌组织中EphA2受体mRNA及蛋白的表达水平。结果:LCM成功地将卵巢癌细胞从卵巢癌冰冻组织切片中分离。RT-PCR结果显示,卵巢癌组织中有不同级别的EphA2受体mRNA表达,mRNA表达与卵巢癌患者的年龄、FIGO分期、组织学类型、组织分化程度以及残余肿瘤大小均无关(P>0.05)。Western blot结果显示EphA2受体表达与mRNA表达水平一致。结论:LCM能够精确分离少量卵巢癌细胞;卵巢癌组织中有不同级别的EphA2受体mRNA及蛋白质表达。
Aim:To investigate the messenger RNA (mRNA) and protein expression of EphA2 receptor in ovarian carcinomas.Methods:Semiquantitative RT-PCR and Western blot analysis assisted by laser capture microdissection(LCM) were used to measure mRNA and protein expressions of EphA2 receptor in 30 cases of ovarian carcinoma.Results:Tumour cells were successfully separated from the surrounding stromal cells by means of LCM.
