奥曲肽对肠源性内毒素肝损害的保护作用

摘要 目的 :探讨奥曲肽对肠源性内毒素肝损害的机理及保护作用。方法 :用 42只SD大鼠制作肠源性内毒素肝损害的动物模型 ,随机分为 7组 ,观察各实验组大鼠血浆内毒素、肿瘤坏死因子 α(TNF α)、ALT、LDH及肝组织病理形态学的变化。结果 :门静脉复流后 ,奥曲肽B2、B3组内毒素、TNF α水平较相应生理盐水A2、A3组明显降低 (P <0 .0 1,P <0 .0 1) ;奥曲肽B2、B3组ALT值较相应生理盐水各组明显下降 (P <0 .0 5、P <0 .0 1) ;奥曲肽B3组LDH值较相应生理盐水A3组亦明显下降 (P <0 .0 5 )。奥曲肽各组的肝脏病理组织学改变较相应生理盐水各组明显减轻。结论 Objective:To study the Protective effect of octreotide on the liver damage induced by intestinal endotoxin. Methods: Rats model of liver damage induced by intestinal endotoxin was produced by blocking portal vein blood flow. Fourty two Sprague Dawley(SD)rats were divided randomly into seven groups. In all groups, plasma endotoxin level, serum TNF αconcentration, ALT, LDH, and the pathological changes of the liver were detected. Results: The results showed that the levels of endotoxin, TNF α, ALT, and LDH were significantly lower in octreotide treated groups of B2 and B3 than those in natural saline treated groups of A2 and A3 after portal vein blood flow was resumed. The pathological changes were alleviated and fine structure of hepatic cells were preserved in octreotide treated group. Conclusion: Occlution portal vein can cause intestinal endotoxemia and liver damage. Otreotide has a portective effect on the lever damage induced by intestinal endotoxin.
