摘要
目的:构建奶水牛乳腺表达人FSHβ基因载体。方法:利用限制性内切酶和T4DNA连接酶方法将转基因构件(水牛β酪蛋白启动子、hFSHβ、SV40PolyA)插入pLXIN载体中,通过转化大肠杆菌,筛选阳性克隆。结果:通过酶切鉴定及测序分析表明pLXIN插入转基因构件。结论:奶水牛乳腺表达人FSHβ基因的载体为:pLXIN-hFSHβ。
Objective To design and construct the mammary gland specific expression vecor of human FSH β(hFSHβ) in buffalo. Method to incorporate the buffalo β casein upstream reglatory sequence and human FSHβ cDNA as well as SV40 early polyA into the pLXIN vector with retriction enzyme and T4 DNA ligase. Results The map of the expression vector by BamHI+HindⅢ showed the positive clone and the incorporated sequence of human FSHβ,which was 100 percent of homologous to the known sequence by DNA sequencing. Conclusion The results showed the vector, pLXIN-hFSHβ, incorporated the human FSHβ cDNA,which was directed by buffalo β-casein promoter.
