摘要
目的探讨阿魏酸钠(sodium femlate,SF)对过氧化氢(hydrogen peroxide,H_2O_2)引起的PC12细胞凋亡保护作用的可能机制。方法在H_2O_2诱导PC12细胞凋亡模型的基础上,采用MTT比色分析检测细胞存活率,RT-PCR检测前列腺凋亡响应基因Par-4(prostate apoptosis response-4,Par-4)和核转录因子-κB p65(nuclear factor-kappaB,NF-KB)mRNA表达,Westem blot检测Par-4和NF-κBp65蛋白表达。结果不同剂量SF预处理1h可提高PC12细胞的存活率,降低Par-4和NF-κBp65的mRNA表达以及蛋白表达。结论SF可剂量依赖性地对抗H_2O_2的神经素毒性作用,其机制可能与凋亡基因Par-4表达有关,与NF-κB的关系尚需进一步探讨。
