摘要
【目的】研究犬脑枪弹伤后神经细胞凋亡机制及c-myc的变化规律。【方法】建立犬颅脑枪弹伤模型,制备不同时期、不同部位脑组织切片。采用末端脱氧核苷酸转移酶介导的生物素脱氧尿嘧啶核苷酸缺口末端标记法(TUNEL)检测脑神经细胞凋亡,用免疫组织化学方法检测c-myc的动态变化。【结果】对照组犬偶见神经细胞凋亡,神经细胞中有微弱c-myc的表达,枪弹伤后凋亡神经细胞于2 h开始增加(P<0.05),24 h达高峰(P<0.01),48 h开始下降。c-myc伤后30 min开始增加(P<0.05),2 h达高峰(P<0.01),6 h开始下降,24 h同对照组。不同区域表达不一致。【结论】犬脑枪弹伤后神经细胞凋亡及c-myc的表达增强,但c-myc先于神经细胞凋亡的表达,在不同时期、不同部位具有时空规律性。神经细胞的凋亡与凋亡保护基因的调节有关。
【Objective】To investigate the mechanism of neuron cell apoptosis and the expression of c-myc in cerebral cortex following craniocerebral gunshot wound in canine.【Methods】Experimental craniocerebral gunshot wound model of canine was set up.Samples of canine brain were taken at different time and areas.Apoptosis cells were detected by terminal deoxynucleotidyl transferease-mediated biotinylated deoxyuridine triphosphate nick and labeling(TUNEL), and c-myc positive neurons were detected by immunohistochemical method.【Results】Weak neuron apoptosis and c-myc were detected in the control group.Apoptosis cells in neurons were found after craniocerebral gunshot wound and increased at 2 h,reached the peak at 24 h,and began to decrease at 48 h.The c-myc positive expression in the above mentioned areas after craniocerebral gunshot wound was increased at 30 min,reached the peak at 2 h,began to decrease at 6 h,and had no difference from the control group at 24 h.The expressions in different areas were different.【Conclusion】Apoptosis and c-myc positive expression in neurons after craniocerebral gunshot wound in canine are increased,but the elevated induction of neuron cells apoptosis appears to be delayed as compared with that of c-myc,and is found to have a temporal and spatial regulation at different time and areas.The neuron cells apoptosis is related to the regulation of anti-apoptosis in neuron.
