摘要
目的:探讨低剂量超氧阴离子自由基(O2-)对小鼠乳腺癌细胞株EMT6的促增殖作用及该作用与NF-κB和细胞周期的关系。方法:黄嘌呤/黄嘌呤氧化酶(XaXO)系统产生的不同剂量O2-直接作用于培养的EMT6细胞,筛选刺激EMT6细胞增殖的最佳剂量:20μmol/LNF-κB抑制剂(PDTC)预处理细胞2h,5.0mU黄嘌呤氧化酶(XO)体系产生的O2-作用EMT6细胞,5.0mVXO体系产生的O2-分别作用于G1、G2/M、S期EMT6细胞,MTT法测定细胞增殖活性。结果:5.0mUXO体系产生的O2-具有明显的促进EMT6细胞生长的作用,此作用可以被PDTC抑制;5.0MuXO体系产生的O2-可以明显的促进G1期EMT6细胞生长。结论:低剂量O2-可诱导EMT6细胞增殖,该过程可能包含依赖NF-kB的信号途径的参与;O2-诱导EMT6细胞增殖时存在细胞周期敏感时相,该敏感时相为G1期。
