摘要
目的通过观察银杏叶提取物注射液(金纳多)对脂多糖(LPS)导致的急性肺损伤大鼠炎症介质释放的影响,从而探讨金纳多对急性肺损伤的保护作用。方法63只大鼠随机分成对照组(尾静脉注射生理盐水)、肺损伤组(尾静脉注射LPS,5mg/kg),治疗组(尾静脉注射LPS5mg/kg+金纳多17.5mg/kg),每组21只。注射后2、6、10h各组分别取7只大鼠获得肺泡灌洗液和血液标本以及肺组织切片,进行检测。结果损伤组中肺系数(0.345±0.031)、肺泡灌洗液总蛋白含量(242±3.97)ng/ml,肺通透指数(2.86±0.85)以及血液中肿瘤坏死因子-α[TNF-α,(2.48±0.12)]g/ml、白介素-6[IL-6,(0.546±0.099)ng/ml]、一氧化氮([NO,(72.9±5.3)mmol/L]均较对照组0.252±0.027,(65.04±2.57)ng/ml,0.75±0.13,(0.88±0.04)ng/ml,(0.078±0.004)ng/ml,(21.8±1.5)mmol/L明显上升(均P<0.05),肺组织病理切片显示肺泡结构破坏严重,而应用金纳多后治疗组各项指标为0.271±0.03、(143.19±13.04)ng/ml,1.67±0.97,(1.64±0.24)ng/ml,(0.18±0.055)ng/ml,(39.7±5.6)mmol/L有明显下降(P<0.05),并且肺泡结构破坏程度减轻。结论应用金纳多LPS导致的实验性肺损伤大鼠的炎症介质释放过程有抑制作用,肺泡破坏减轻,提示银杏叶提取物注射液可以减轻大鼠实验性肺损伤的程度,可以保护肺组织。
Objective To investigate the protective effect of injection of ginkgo biloba leaves extract on acute lung injury in rats Methods Sixty three Wistar rats were randomized into three groups(n=21 in each group): group C (control group) in which rats were injected with normal saline into vena caudalis, group I (lung injury group),in which rats were injected with lipopolysaccharide (LPS, 5mg/kg) and group T (treatment group),in which rats were injected with LPS 5mg/kg and extract of ginkgo biloba 17.5mg/kg into vena caudalis). After 2h, 6h, 10h of injection the blood samples and bronchoalveolar lavage fluid were collected, and pathological sections from lung in 7 rats from every group were examined. Results In group I, the levels of lung coefficient 0.345±0.031, the protein concentration of BALF(242±3.97)ng/ml, the lung penetrating index(LPI)(2.86±0.85)and the TNF-α(2.48±0.12)ng/ml, IL-6(0.546±0.099)ng/ml, NO(72.9±5.3)mmol/L levels in blood samples were all higher than those in group C(0.252±0.027,65.04±2.57)ng/ml,(0.75±0.13,0.88±0.04)ng/ml,(0.078±0.004)ng/ml,(21.8±1.5)mmol/L, respectively. In group I, the degree of disorganization of lung alveolus was very serve, and in group T the corresponding parameters 0.271±0.03,(143.19±13.04)ng/ml,1.67±0.97,(1.64±0.24)ng/ml,(0.18±0.055)ng/ml,(39.7±5.6)mmol/L, respectively were significantly lower. Conclusion Ginkgo biloba extract can suppress the mediators of inflammation of the ALI induced by lipopolysaccharide, and it can alleviate disorganization of lung alveolus.
