心房颤动病人心房肌肌浆网Ca^(2+)泵和Ca^(2+)释放通道的变化

摘要 目的探讨心房颤动病人心房肌肌浆网Ca2+泵(sarcoplasmic reticulum Ca2 +ATPase,SERCA2a)及钙释放通道2型雷尼丁受体(type 2 ryanodine receptor,RYR2)mRNA表达的变化。方法39例风湿心脏病二尖瓣关闭不全接受外科手术者,分为3组,窦性心律组13例,心房颤动持续小于6个月组11例,心房颤动持续超过6个月组15例。手术时取右心房肌约100mg,用逆转录-聚合酶链反应检测心房肌SERCA2a和RYR2的mRNA表达。结果心房颤动者肌浆网SERCA2a、RYR2的mRNA较窦性心律者下调,而且下调随心房颤动持续时间延长而明显。结论心房颤动病人SERCA2a、RYR2的mRNA下调,提示肌浆网SERCA2a、RYR2与心房颤动的发生和维持有关。 Objectives To investigate mRNA expression changes of sarcoplasmic reticulum Ca2+ ATPase (SERCA2a) and calcium releasing channel type 2 ryanodine receptor (RYR2) of atrium in patients with atrial fibrillation. Methods 39 patients with mitral valve disease were divided into three groups, namely sinus rhythm group, atrial fibrillation less than 6 month group and atrial fibrillation more than 6 month group. during the open heart surgery,100 mg of tissues were obtained from the right atrium respectively to detect mRNA expressions of SERCA2a and RYR2 by RT-PCR. Results Down regulation of SERCA2a and RYR2 mRNA expressions were observed, the longer the duration of atrial fibrillation, the more obvious the down regulation. Conclusions The down regulation of SERCA2a and RYR2 may have correlation with the initiation and maintenance of atrial fibrillation.
