摘要
目的 检测颞叶癫痫大鼠模型中Robo3的动态表达,探究其在颞叶癫痫发病机制中的作用.方法 雄性SD大鼠36只随机分为6组(n=6):1个对照组和5个实验组,实验组腹腔注射氯化锂-匹罗卡品(LiCl-PILO)建立颞叶癫痫大鼠模型,对照组注射等量生理盐水,5个实验组分别于致痫后1d(急性期),7、14 d(静止期),30、60 d(慢性期).取海马组织为研究对象,并与对照组比较.荧光定量PCR检测Robo3 mRNA的表达变化及规律,Western blot检测Robo3蛋白在大鼠海马组织中的表达变化及规律,并进行统计学分析.结果 实验组造模成功率为86.7%.荧光定量PCR结果显示:与对照组相比,致痫后急性期的大鼠海马组织中的Robo3 mRNA表达无明显变化,差异无统计学意义(P>0.05),在静止期和慢性期大鼠海马组织中的Robo3 mRNA表达降低,差异有统计学意义(P<0.05),并在静止期达到最低表达.Western blot结果显示:与对照组相比,致痫后急性期大鼠海马组织中Robo3蛋白表达无明显变化,差异无统计学意义(P>0.05),但在静止期和慢性期Robo3蛋白表达降低(P<0.05),在慢性期达到最低值.结论 Robo3在颞叶癫痫大鼠模型的静止期和慢性期的表达水平显著降低,并在慢性期达到最低值,提示Robo3可能参与颞叶癫痫的发生发展有密切相关.
Objective To characterize the dynamic expression of Rob03 in the rat model of temporal lobe epilepsy(TLE),and assess the potential contribution of Robo3 to epileptogenesis.Methods Male Sprague-Dawley(SD)rats were randomly divided into the control group(n=6)and the experimental groups(n=30,6 per group).The experimental gmups were injected intraperitoneally(i.p.)with an aqueous solution of lithium-pilocarpine,and sacrificed at different time points(1,7,1 4,30 and 60 days)following the seizure.The control group was i.p.with 0.9%sodium chloride instead of pilocarpine.Quantitative real-time PCR were used to detected the mRNA expression of Robo3 and Western bolt were used to detected the protein expression of Rob03.Results Quanmative real-time PCR showed fhat che expression of Rob03 were signi6cantly lower in the mt temporal lobe tissues of the latent and the chronic period group as compared with the controls(P<0.05),but no significant difkrences were identified between the acute period group and the controls(P>0.05).Western blot showed that the pmtein expression of Rob03 were significantly lower in the mt temporal lobe tissues of the latent and the chronic period group as compared with the controls(P<0.05),no signi6cant differences were identi“ed between the acute period gmup and the contIDls(P>0.05).Conclusions Rob03 may be inv01Ved in the pathogenesis of temporal lobe epilepsy.
