摘要
目的:研究柴胡总皂苷对小鼠肝脏葡萄糖醛酸转移酶(UGT)1A1和硫酸转移酶(SULT)1A1活性及mRNA表达的影响.方法:将30只小鼠随机分为3组,雌雄各半,分别为纯水组(10 mL/kg)、柴胡总皂苷(TSS)高、低剂量组(给药剂量分别为150 mg/kg、50 mg/kg),连续灌胃7 d,每天2次.用差速离心法分离肝微粒体和胞质液;以分光光度法检测UGT1A1、SULT1A1活性;以荧光定量PCR法检测UGT1A1和SULT1A1 mRNA在肝脏中的表达.结果:柴胡总皂苷高、低剂量组小鼠肝脏UGT1A1活性与纯水组比较差异无统计学意义(P>0.05);柴胡总皂苷高、低剂量组小鼠SULT1A1活性均高于纯水组,差异有统计学意义(P<0.05);SULT1A1基因表达测定中,柴胡总皂苷高剂量组高于纯水组,差异有统计学意义(P<0.05).结论:柴胡总皂苷能明显诱导小鼠肝脏SULT1A1酶活性和mRNA的表达水平,对小鼠肝脏UGT1A1活性没有影响.
Objective:To study the effect of total saponins of radix bupleuri on activities and mRNA expressions of uridine glucuronosyl transferase(UGT)1A1 and sulfate transferase(SULT)1A1 in livers of mice.Methods:30 mice were randomly divided into 3 groups,half male and half female,a pure water group(10 mL/kg),a high-dose group of total saponins of radix bupleuri(TSS)(150 mg/kg),a low-dose group of total saponins of radix bupleuri(50 mg/kg),and continuous gavage was performed for 7 days,twice a day.The liver microsomes and cytoplasmic fluid were separated by differential centrifugation.The activities of UGT1A1 and SULT1A1 were detected by spectrophotometry.The mRNA expressions of UGT1A1 and SULT1A1 in livers were detected by real-time PCR.Results:There was no statistically significant difference in the activity of UGT1A1 in livers of mice between the high-dose and low-dose groups of total saponins of radix bupleuri and the pure water group(P>0.05).The activity of SULT1A1 in livers of mice in the high-dose and low-dose groups of total saponins of radix bupleuri were higher than that in the pure water group,and the difference was statistically significant(P<0.05).The expression of SULT1A1 gene in the high-dose group of total saponins of radix bupleuri was higher than that in the pure water group,and the difference was statistically significant(P<0.05).Conclusion:Total saponins of radix bupleuri can significantly induce the activity and mRNA expression of SULT1A1 enzyme in livers of mice,and have no effect on the activity of UGT1A1 in livers of mice.
