摘要
目的研究SUMO特异性蛋白酶(SENPs)在原发性肝癌组织中的表达及意义。方法采用免疫组织化学染色法检测SENPs在40例原发性肝细胞癌组织芯片及30例正常对照肝组织中的表达,做统计学比较;并分析SENPs在肝细胞癌组织中表达情况与肝细胞癌临床病理特征间的关系。结果肝细胞癌中SENP1、SENP2和SENP3的表达率显著低于正常肝组织。SENP6、SENP7蛋白阳性表达与肝细胞癌的埃德蒙森分级(低级别或高级别)有关(P<0.05),随着级别升高,两者表达率显著升高。SENP1、2、3、5在肝细胞癌中表达率与患者性别、年龄、肿瘤大小及组织学分级并无显著相关性(P>0.05)。结论SENP1、SENP2和SENP3可能是肝细胞癌变过程中的关键因子;SENP6、SENP7可能在肝细胞癌演进过程发挥重要作用;提示SENP1、SENP2、SENP3、SENP6、SENP7可能成为研究肝细胞癌靶向治疗的潜在靶点。
Objective To study the expression and significance of SENPs in primary hepatocellular carcinoma(HCC).Methods Immunohistochemical staining was used to assess the expression of SENPs in 40 HCC tissue microarrays;and 30 normal liver tissues were selected as control group.The expression of SENPs in HCC and its correlations with clinicopathological features were analyzed.Results The expression rates of SENP1,SENP2 and SENP3 in HCC were significantly lower than those in control group.The positive expression of SENP6 and SENP 7 proteins was correlated to Edmonson grade(low or high grade)of HCC(P<0.05).With the increase of grade,the expression rate of the two genes was significantly increased.But the expression rates of SENP1,SENP2,SENP3 and SENP5 were not correlated to sex,age,tumor size or histological grade.Conclusion SENP1,SENP2 and SENP3 may be the key factors in hepatocellular carcinogenesis.SENP6 and SENP7 may play important roles in the progression of hepatocellular carcinoma.These results suggest that SENP1,SENP2,SENP3,SENP6 and SENP7 may be potential therapeutic targets for HCC.
