摘要
为探究结直肠癌患者体内异常代谢通路及发现肿瘤代谢标志物,将结直肠癌患者22例纳入CRC组,痔患者16例纳入对照组。利用超高效液相色谱串联飞行时间质谱(UPLC-TOF-MS)的定性鉴别能力,对结直肠癌患者与正常人组织样本中的代谢物进行分析,通过主成分分析和正交偏最小二乘分析进行统计学描述,结合人类代谢组数据库进行筛选。结果显示,最终在正负模式下共筛选出3种具有显著性差异且与结直肠癌有密切关联的代谢物,分别为氨基乳糖苷、核黄素环-4’,5’-磷酸盐和琥珀酸。结果表明,该方法可对结直肠癌组织样本中的代谢物进行快速检测,筛选出的代谢标志物可以对结直肠癌的诊断、分期以及发病机制研究提供重要帮助。
In order to investigate the abnormal metabolic pathway of colorectal cancer(CRC)patients,and to seek the tumor metabolic markers,authors enrolled 22 patients with colorectal cancer into CRC group,and 16 patients with hemorrhoids into control group,then by using ultra performance liquid chromatography time of flight mass spectrometry(UPLC-TOF-MS)qualitative differential capacity analysed the metabolites of patients and health person samples,through main component analysis and orthogonal least square method descripted them statistically,and combined with human metabolimeics database to sreen them.As results,finally according to positive-negative mode the totalled 3 types of metabolites were screened out which have significant difference each other,and close relation with colorectal cancer:amino lactoglycoside,riboflavin ring-4’,5’-phosphated and succinic acid.Results show that above methods can fastly detect the metabolites in tissue samples of colorectal cancer,and screen out metabolic marketrs,so that make out diagnosis,staging,and provide help to pathogenesis mechanism research.
