摘要
目的探讨内质网应激PERK/eIF-2a通路与肝癌发生发展的关系。方法选取2015年1月-2017年1月本院收治的原发性肝癌患者80例,联合使用吉西他滨与顺铂治疗2个疗程。采用免疫组织化学法检测肝癌患者癌组织中PERK和eIF-2a蛋白的表达;采用Real-Time PCR法和Western blot法检测肝癌组织中PERK、eIF-2a、ATF6、CHOP、XBP-1和Caspase12 mRNA和蛋白的表达。结果对原发性肝癌患者联合使用吉西他滨与顺铂治疗,化疗后原发性肝癌组织中PERK和eIF-2a蛋白免疫组化检测显示呈高表达水平,且化疗前、后PERK和e IF-2a的表达强度差异有统计学意义(P<0.05)。化疗后肝癌组织中PERK、eIF-2a、ATF6、CHOP、XBP-1和Caspase12 mRNA和蛋白表达含量均显著升高,差异有统计学意义(P<0.05)。结论吉西他滨联合顺铂对原发性肝癌的有一定的治疗作用,且内质网应激PERK/eIF-2a通路在此过程中发挥重要的作用。
Objective To investigate the relationship between PERK/eIF-2a pathway and the development of hepatocellular carcinoma(HCC).Methods A total of 80 patients with primary liver cancer treated in our hospital from January 2015 to January 2017 were selected and treated with gemcitabine and cisplatin for 2 courses.Immunohistochemical method was used to detect the expression of PERK and eIF-2a protein in cancer tissues of patients with hepatocellular carcinoma.Real-time PCR and Western Blot were used to detect the expression of PERK,eIF-2a,ATF6,CHOP,XBP-1 and Caspase12 mRNA and protein in HCC.Results The combination of gemcitabine and cisplatin was used in patients with primary liver cancer.Immunohistochemical detection of PERK and eIF-2a proteins in primary liver cancer tissues after chemotherapy showed high expression levels,and the expression intensity of PERK and eIF-2a before and after chemotherapy was significant higher(P<0.05).The PERK and eIF-2a were significantly higher in primary hepatocellular carcinoma after chemotherapy.The expressions of PERK,eIF-2 a,ATF6,CHOP,XBP-1 and Caspase12 mRNA and protein in hepatocellular carcinoma were significantly increased after chemotherapy,there was statistically difference(P<0.05).Conclusion Gemcitabine combined with cisplatin has a certain therapeutic effect on primary liver cancer.And the endoplasmic reticulum stress PERK/eIF-2a pathway plays an important role in this process.
