Transcriptome Sequencing Revealed Differences in Gene Expression Between Male and Female Venom Glands of Wolf Spider(Lycosa sp.)

摘要 蜘蛛毒液蕴藏着大量的生物活性物质,在生物医药、新型杀虫剂开发等领域具有广阔的应用前景。本研究旨在分析狼蛛(Lycosa sp.)毒腺中基因表达的性别差异,为蜘蛛毒液分类开发利用提供理论依据。本研究采用高通量测序技术对不同性别狼蛛(Lycosa sp.)的毒腺进行了转录组测序,雌性组获得44394136条原始Reads,雄性组获得40502072条原始Reads,de novo拼装获得78451个Unigenes。利用生物信息学方法进行数据分析,这些Unigenes经注释共比对到11个近缘物种,其中6个为蜘蛛物种,且同属狼蛛科(Lycosidae)。两性毒腺共鉴定得到403个差异Unigenes,与雄性狼蛛相比,雌性中上调基因78个,下调基因325个。GO富集显示上调基因多与毒素活性(Toxin activity)、防御反应(Defense response)、细胞杀伤(Cell killing)、细胞溶解(Cytolysis)等通路相关,而下调基因多富集在RNA稳定性相关通路。研究结果显示狼蛛毒腺中基因表达具有性别差异,推测雌性狼蛛(Lycosa sp.)可能更具攻击性或毒性更强。本研究为狼蛛(Lycosa sp.)毒液的深入研究提供了基础资料,为蜘蛛毒液进化研究提供了原始数据。 Spider venom contains a large number of bioactive substances,which has a wide range of application prospects in biological medicine,novel pesticide development and other fields.This study aimed at analysing the gender differences in the expression of the venom gland of wolf spiders(Lycosa sp.)and provided a theoretical basis for the classified utilization of spider venom.In this study,high-throughput sequencing technology was used to sequence the venom gland transcriptome of different gender wolf spiders(Lycosa sp.).44394136 original reads were generated in female individuals,40502072 original reads were obtained in male individuals,and 78451 unigenes were assembled by de novo.The bioinformatics method was used to analyze data,and a total of 11 related species was compared,6 of which are spider species,and all belong to Lycosidae.A total of 403 differential unigenes was identified in the venom glands of both sexes.Compared with males,there were 78 up-regulated genes and 325 down-regulated genes.GO enrichment showed that up-regulated genes were mainly associated with toxin activity,defense response,cytotoxicity and cytolysis,while down-regulated genes were mainly associated with RNA stability.This study showed some differences in venom gland expression between males and females,and we hypothesized that female wolf spiders(Lycosa sp.)may be more aggressive or toxic.This study provides the preliminary data for the further study of wolf spider(Lycosa sp.)venom and the original data for the study of the evolution of spider venom.
