白藜芦醇联合5-氨基水杨酸对脂多糖诱导的HT-29细胞损伤模型紧密连接蛋白表达的影响 被引量:1

摘要 目的:探究白藜芦醇(RSV)联合5-氨基水杨酸(5-ASA)对脂多糖(LPS)诱导的HT-29细胞紧密连接蛋白损伤的影响及其机制。方法:用100μg/mL的LPS刺激人结肠腺癌细胞HT-29,分为LPS组、5-ASA组、RSV组和RSV+5-ASA组,同时设对照组。用CCK-8法检测细胞活力,western blotting法检测细胞occludin、claudin-1、ZO-1、Notch1、Hes1蛋白表达,透射电镜观察细胞的紧密连接结构。结果:与对照组比较,LPS组细胞紧密连接蛋白occludin、claudin-1、ZO-1表达水平降低,Notch1、Hes1蛋白表达水平升高(均P<0.05),且紧密连接结构模糊,呈连续性中断。与LPS组比较,5-ASA组、RSV组和RSV+5-ASA组细胞紧密连接蛋白occludin、claudin-1、ZO-1表达上调,Notch1、Hes1蛋白表达下调(均P<0.05),紧密连接结构致密,其中RSV+5-ASA组最为显著(P<0.05)。结论:RSV与5-ASA联用可上调HT-29细胞损伤模型紧密连接蛋白occludin、claudin-1、ZO-1的表达,该过程可能与抑制Notch1信号通路活性有关。 Objective:To explore the effect and mechanism of resveratrol(RSV)combined with 5-aminosalicylic acid(5-ASA)on tight junction protein expression in lipopolysaccharide(LPS)-induced HT-29 cells.Methods:Human colorectal adenocarcinoma cells HT-29 were stimulated with 100μg/mL LPS and divided into LPS group,5-ASA group,RSV group,RSV+5-ASA group,and control group.Cell viability was determined by CCK-8 method.The protein expressions of occludin,claudin-1,ZO-1,Notch1 and Hes1 were detected by western blotting.Transmission electron microscopy(TEM)was used to observe the tight junction structure of cells.Results:Compared with the control group,the protein expressions of tight junction proteins occludin,claudin-1 and ZO-1 decreased in LPS group,while the protein expressions of Notch1 and Hes1 increased(all P<0.05),and the tight junction structure was blurred,presenting continuity interruption.Compared with LPS group,the expressions of tight junction proteins occludin,claudin-1 and ZO-1 in 5-ASA group,RSV group and RSV+5-ASA group were up-regulated,the protein expressions of Notch1 and Hes1 were down-regulated(all P<0.05),and the tight junction structure was dense.RSV+5-ASA group was the most significant(P<0.05).Conclusion:RSV combined with 5-ASA can up-regulate the expressions of tight junction proteins occludin,claudin-1 and ZO-1 in HT-29 cell injury model,which may be related to the inhibition of Notch1 signaling pathway activity.
